Mechanism of Action.
Selank is a synthetic heptapeptide consisting of tuftsin (Thr-Lys-Pro-Arg), a naturally occurring immunostimulatory tetrapeptide, extended with a stabilizing Pro-Gly-Pro tripeptide. Its anxiolytic mechanism involves modulation of the GABAergic system, increasing GABA receptor sensitivity without directly binding GABA receptors (unlike benzodiazepines). Selank influences the balance of monoamine neurotransmitters, affecting serotonin metabolism and dopamine activity. It inhibits the enzyme enkephalinase, increasing endogenous enkephalin levels which contribute to mood regulation and stress resilience. Additionally, selank modulates the expression of interleukin-6 and affects the balance of Th1/Th2 immune responses through its tuftsin-derived immunomodulatory activity.
Research Summary.
Selank has been studied extensively in Russia for generalized anxiety disorder (GAD) and neurasthenia. Clinical studies demonstrated anxiolytic effects comparable to medazepam (a benzodiazepine) without the sedation, cognitive impairment, or dependence potential. Research shows selank influences the expression of 36 genes related to GABAergic and serotonergic neurotransmission. Studies in patients with anxiety disorders showed significant reductions in Hamilton Anxiety Rating Scale scores. The immune-modulating properties of selank have been studied in the context of viral infections, showing enhanced NK cell activity and modulation of inflammatory cytokines. Selank has also demonstrated nootropic properties, improving memory and learning in animal models, though these effects are less pronounced than those of semax.
Side Effects & Safety.
Selank has demonstrated a very clean safety profile in clinical research and pharmaceutical use in Russia. No serious adverse events, sedation, cognitive impairment, withdrawal symptoms, or dependence have been reported. Mild nasal irritation may occur with intranasal use. The absence of GABA receptor agonism means selank avoids the dependency and tolerance issues associated with benzodiazepines. It does not impair motor function or reaction time. As with semax, the formal Western safety dataset is limited, with most data coming from Russian clinical experience.
